Objectives:
In vitro culture

  • experimental embryology and somatic embryogenesis - plant regeneration by induction of somatic embryos from different explants, effect of light spectral properties on somatic embryogenesis,
  • androgenesis - regeneration of DH plants and homozygous lines from isolated microspores and anther culture (wheat, rye, triticale, and barley) for breeding progress
  • cryopreservation in in vitro cultures for Gen Bank conservation,
  • transgenic plants, plant tissue transformation for research study and industrial use (particle gun, Agrobacterium, PEG),
  • RAPDs and numerical taxonomy - analysis of genetic distance and diversity to establish the stability of lines and their relationship.

Biosafety activity www.ihar.edu.pl/gene_bank/GMO

  • creation of GMP diagnostic laboratory for quantitative and qualitative GMO detection,
  • rising public awareness on GMP as a member of International "GMP-Network",
  • supporting the GMO law implementation in Poland,
  • contribution to the National Biosafety Framework of Poland,
  • gene expression study.

Laboratory of Cytogenetics
Jerzy Macewicz, D.Sc.

Objectives:
Variation in the 3n subaleurone protein tissue of barley

  • protein content in doubled haploids and diploid lines,
  • inter- and intra- spike variation in the 3n subaleurone protein tissue,
  • study of the inter- and intra spike variation of protein content in the 3n subaleurone layer,
  • histochemical test for protein quantity assessment in single seeds,
  • inheritance of subaleurone proteins in selection for brewery types of barley,
  • the effect of endosperm protein content and phenotypic characteristics of sublines.

Biomechanics of crop plants

  • mechanical properties of stem in cereals and pea in relation to lodging resistance,
  • heritability and genetic markers of stem mechanical traits,
  • fracture properties of seeds in relation to technological quality,
  • photomorphogenesis and plant biomechanics and architecture.

Role of cell wall and cell wall proteins in control of plant growth

  • acid growth, cell wall rheology in relation to shoot elongation rate,
  • expansin expression and activity,
  • photomodulation of growth rate and cell wall proteins expression,
  • cell wall modifications in low temperature hardening of rape plants.

Cellulose biosynthesis

  • expression of cellulose synthase subunits,
  • photocontrol of cellulose synthesis,
  • cell wall structural mutations.

Morphological traits of the ear and the grain yield

  • morphogenesis of the ear, spikelets and florets in cereals,
  • effect of duration of particular ear developmental phases on grain number,
  • ear morphogenesis analysis for in in vitro microspore and anther cultures.
